Associated Literature (AI-based literature mining, Beta-version)

Plant U-box E3 Ubiquitin Ligase NOP1; NOP1 has an essential role in schizogenous intercellular space formation in Marchantia polymorpha.NOP1 encodes a Plant U-box (PUB) E3 ubiquitin ligase carrying tandem ARMADILLO (ARM) repeats in the C terminus. An in vitro ubiquitination assay indicated that the NOP1 protein possesses E3 ubiquitin ligase activity in a U-box-dependent manner. Confocal microscopy and biochemical analysis showed that NOP1 was localized to the plasma membrane. (from nomenclature)

Sugano, S. S., et al. (2018) · PLoS ONE  research experimental subject
Targeted to assess editing efficiency via the easily scored transparent-thallus (air-chamber-less) phenotype; 87.5% of 32 T1 plants were mutated. Used to test guide length (17nt or fewer dramatically less efficient) and to induce 0.5/1.5/4.5kb deletions with paired gRNAs; no off-target mutations.
Kopischke, S., et al. (2017) · Plant Methods  research experimental subject
Selected as the TALEN knockout target because its mutant lacks air chamber formation (an easily traceable phenotype); both TALEN approaches achieved >20% targeting efficiency with deletions of 4 nt to ~2000 nt across 22 sequenced T1 lines. Two mutants carried in-frame indels revealing novel Mpnop1-related phenotypes, with stable inheritance across generations.
